{"id":13862,"date":"2021-12-09T16:45:31","date_gmt":"2021-12-09T22:45:31","guid":{"rendered":"https:\/\/www.coleparmer.com\/blog\/?p=13862"},"modified":"2021-12-09T16:45:31","modified_gmt":"2021-12-09T22:45:31","slug":"rapid-results-when-testing-for-aspergillus","status":"publish","type":"post","link":"https:\/\/www.coleparmer.com\/blog\/rapid-results-when-testing-for-aspergillus\/","title":{"rendered":"How to Achieve Rapid Results When Testing for Aspergillus"},"content":{"rendered":"<h2>A new detection kit can expedite your qPCR process.<\/h2>\n<p>If you are testing for <em>Aspergillus<\/em> in cannabis and other agricultural products, you can obtain rapid and accurate test results with high specificity by using the new Spex CertiPrep\u00ae 2-Color <em>Aspergillus<\/em> Detection Kit for qPCR. The kit is designed to detect the presence of the four most common pathogenic strains of <em>Aspergillus<\/em> DNA: <em>niger<\/em>, <em>terreus<\/em>, <em>flavus<\/em>, and <em>fumigatus<\/em> DNA in cannabis species flower, stem and\/or leaf tissue(s).<\/p>\n<h2>What\u2019s in the kit?<\/h2>\n<p>Components of the kit include 2 x <em>Aspergillus<\/em> detection Master Mix, <em>Aspergillus<\/em> positive control, internal control, yellow dye (20 x), DNase\/RNase\/Proteinase free water and ROX. For machines that lack a HEX filter, VIC can be interchangeably used.<\/p>\n<h2>How it works<\/h2>\n<p>The internal control ensures the sample is free of PCR inhibitors that would lead to false negative results. To help distinguish the Master Mix from the template DNA and from the final product during sample preparation, an inert yellow dye is provided to label the template prior to adding to the Master Mix which contains an inert blue dye. When mixed the resulting solution is green, confirming the presence of all components. The kit works with any qPCR system that supports the two-color channels. It is very sensitive and can detect down to 10 copies of <em>Aspergillus<\/em> DNA.<\/p>\n<h2>Protocol<\/h2>\n<p>Once all components in the kit are thawed in ice, the tubes are mixed by inversion to ensure homogeneity. Before first use, ROX is added to the 2X Aspergillus Detection Master Mix to ensure qPCR machine compatibility. Afterwards, 10 \u03bcL of the 2X Aspergillus Detection Master Mix is added to designated wells of a qPCR plate. Then, in a separate 1.5 mL tube the following components are mixed:<\/p>\n<table>\n<tbody>\n<tr>\n<td width=\"192\"><strong>Component<\/strong><\/td>\n<td width=\"192\"><strong>Volume (10 \u00b5L)<\/strong><\/td>\n<\/tr>\n<tr>\n<td width=\"192\">20 X Yellow Dye<\/td>\n<td width=\"192\">1 \u00b5L<\/td>\n<\/tr>\n<tr>\n<td width=\"192\">DNA Template*<\/td>\n<td width=\"192\">1\u20136 \u00b5L<\/td>\n<\/tr>\n<tr>\n<td width=\"192\">Internal Control Template<\/td>\n<td width=\"192\">2 \u00b5L<\/td>\n<\/tr>\n<tr>\n<td width=\"192\">Water<\/td>\n<td width=\"192\">up to 10 \u00b5L<\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n<ul>\n<li>For positive control replace DNA template with 2 \u03bcL of Aspergillus Positive Control Template.
